The skin constitutes the body’s outermost barrier, and keratinocytes have a central role in the wound healing process by contributing to the restoration of epithelial tissue. The aim of this study was to investigate how LPS, as a model of bacterial infection, affects wound healing in HaCaT cells through analysis of cell migration, cell viability, and cytokine response. The study was conducted as an experimental in vitro study in which cell migration was analyzed using scratch assay, cell viability using MTT assay, and cytokine quantification using sandwich ELISA. The results showed that the highest concentration of LPS (10 µg/ml) was associated with the smallest reduction in the wound gap over 24 hours, whereas the control group showed the greatest reduction. Cell viability remained relatively unchanged between all treatment groups and was close to the control level. IL-6 could only be detected at the highest LPS concentration and only in two out of three experiments, whereas TNF-α could be detected in all samples and showed higher levels at 10 µg/ml LPS. In conclusion, the results indicate that LPS primarily affects the migratory capacity of HaCaT cells rather than their viability, and that higher concentrations of LPS may induce a limited but measurable inflammatory response
