Small RNA Modulation of TP53 and ADAR Expression : A Comparative Study in HEK-293T and HeLa Cell Lines

Downregulation of gene expression is an essential mechanism for regulating cellular activity. This process is particularly important in the study of diseases and in the development of novel therapeutic strategies. The aim of this study was to identify novel small RNA molecules capable of silencing the target genes Tumor Protein 53 (TP53) and Adenosine Deaminase Acting on RNA (ADAR), and to evaluate their potential therapeutic relevance. Novel small RNAs were transfected into HEK-293T and HeLa cells. Relative gene expression levels were determined using real-time quantitative polymerase chain reaction (qPCR). The results demonstrated suppression of TP53 expression in both cell lines, although the magnitude of downregulation varied depending on the small RNA molecule used. Analysis of ADAR expression revealed downregulation in HeLa cells. These findings indicate that the transfection affected the gene expression level of the target genes, with an exception for ADAR in HEK-293T cells.
