The effect of hemolysis in serum on clinical chemistry parameters in veterinary medicine

Hemolysis is a common preanalytical source of error that may affect the analysis of diagnostic biomarkers in serum samples. The aim of the study was to determine the highest acceptable degree of hemolysis for each of the 30 different biomarkers in equine, canine and feline serum on DxC 70 AU (Beckman Coulter). Using 18 equine, 22 canine- and 15 feline samples with different additions of prepared hemolysates, duplicates of each were analyzed. The interference was evaluated based on the percentage deviation from the sample without addition of hemolysate. A percentage deviation <3% was used as the acceptable limit. Albumin, alkaline phosphatase, aspartate aminotransferase, glucose, calcium, chloride, creatinine, creatine kinase, sodium, serum amyloid A, triglycerides and urea, can be analyzed in serum samples with free hemoglobin concentrations ut to 5g/L while maintaining clinical relevance. Total bilirubin, fructosamine and zinc should only be analyzed in serum samples without visible hemolysis. This applies to all species. Generally, phosphate, bile acids, gamma-glutamyl transferase, glutamate dehydrogenase, iron, potassium, cholesterol, magnesium, total protein and unsaturated iron-binding capacity, could only tolerate hemolysis level between 1g/L and 4g/L. C-reactive protein and cystatin C was only analyzed on canine serum, they can both be analyzed with a hemolysis level up to 5g/L. On canine and feline serum only, alanine aminotransferase, amylase and lipase were analyzed.
